as long as the mildew is taken care of you are fine to clone..
perlite has nothing to do with anything stem/stalk related.. perlite is used for aeration.. it is important as a soil additive as it allows for proper drainage, water retention, and aeration.. but it has nothing at all to do with how thick your stem should be...

as far as pruning and removing leaves goes, i highly recommend not doing it... ill state my case and leave it at...
i dont know brands or wattages of cfls... BUT... http://www.walmart.com/catalog/product.do?product_id=5650616 here is a 6 pack.. for 15.. which is cheap as hell i think.. if i remember right cfls are kinda expensive.. but basically here is the rule of thumb... the smaller the bulbs wattages...
it takes a week or so for the actual knot to form..
the plant will heal and be able to be super cropped before the knot forms...
but the "scar" tissue takes a little longer to form... or rather to completely scar up.. if that makes sense..

if you are using bloom nutes that have N in it its fine... n is not utilized as much during flowering as it is during veg.. so your plant doesnt need the levels of nitrogen during flowering as it...

nitrogen isnt utilized during flowering by the plant.. or at least it uses much much less than it does during veg.. if you are using a bloom nute formula that has nitrogen in it tho, its ok... it wont affect anything.. just keep it all looking nice...

you dont need huge spaces for roots... as long as you can supply the plant with plenty of water and nutes (via a drip system or something along those lines) then it will be fine and can grow quite large...

i added a 4000w flower room, another ebb and gro system, another water chiller (all this in the flower room)...
