yea I am ready to spawn to poo. I know about g2g but have to be more sterile and syringes only last so long so I figured I'd just use them up. I will prob g2g any strains I want to keep around and then just print anything else for storage. A well stored print can last over 5-10 years.
Ok. Someguy. When are you going to do an LC (liquid culture)? This will cut down on your costs considerably.
Since you're only going to do this for a run or two. You probably wouldn't want to waste any time on an LC.
wow... I cannot believe that taking a tissue sample from a single fruit is not an isolation. That's hard to wrap my head around. Ok so say you have 3 "phenos" (spores) since the mycelium can connect as a single network and then fruit, it's possible for the fruit to contain genetic material from multiple "phenos" or spores? Fuckin crazy.No an LC can still be multispore, but it will be maybe two or 3 variants, yeay they are weird, they grow into each other. and even useing a wedge from well inside the stem is no gaurantee of an isolate. That I'm afraid can only done with certainty on agar plates. Haveing said that, L/C's are realy easy to make, and a good way of storeing DNA for a while. I posted a recipe useing coffee rinds and sugar...

And liquid culture is cool, but beyond my needs right now. Only made 7 quarts and 2 3lb bags and I think I'm already gonna have too much spawn.
Also this may seem like a dumb question, but under a scope could one physically manipulate the spores to isolate a single spore for inoculation? My intiution tells me they are too small to manipulate but shit they can fertilize eggs by injecting them and crazy shit so who knows.
And picked this up to expand my knowledge, great book so far.
