You can only really tell by looking at your trichomes buddy. Get a jewler's loupe or something similar and if you have white trichomes with some ambers mixed in you're good to chop. Some wait for more amber some less, that's up to you...

Allbr they do not start flowering immediately, most start flowering 3-4 weeks in. The point of an autoflower is that you don't have to swap light schedules, and they flower themselves. It has nothing to do with flowering immediately.

Have you checked the PH with a pen? RO water can come out in the 5-7 range. Typically it's closer to 6.5.. but it's possible it's off.

It looks like a small calmag deficiency that may have taken care of itself as the new growth looks OK.. It doesn't look like any kind of nute burn I've ever seen. Keep your PH and RH right and keep nutes at half strength till they get bigger
