That is not a pollen sack, just an enlarged calyx. Common to see large calyx on some species at the node. Only the head of the trichrome turns amber. The only way to see them is with a scope.

When I cut a sample for trics, I cut a very small, I mean like a 2-4cm size sample. Usually something flat like a sugar leaf and small enough to fit in the plastic circle of a digital scope
