Weird spot on the leaf, when should i transplant?

I'm a first time indoor grower (finally got my card) I only plan on growing 2 at a time.

Im growing under cool white florecent bulbs right now, I will switch to HPS later on.

My questions are....

1. What is the yellow spot on that one leaf? An should i just get rid of the plant so the other one doesnt get it?

2. How much longer should i wait to transplant them into bigger separate containers? I put a pack of cigarettes next to them for comparison (i dont smoke cigarettes, my roommate does, but not around my plants..i know its bad for them)

need 2 go ahead and seperate the plants and if ure groen in soil do not use mg moisture control or any chemical nuted potting soil . mg organic and alaska fish emulsion is ok if u have 2 shop at lowes , if u have a local hydro store theyll set u up with the good stuff :leaf:
