I'm aware of when to harvest in terms of looking for trichome cloudiness and amber coloring, but in order to view my buds in the microscope I would need them off the branch. Does everyone else just snip a tiny amount for a sample or just view the buds while still on the plant? The reason I ask is because my microscope is not the highest quality but still has a 40x zoom. Any advice/steps for obtaining a sample to view would be appreciated.

I sure do - I usually snip off a leaf that is covered with trichomes that's part of a bud and slip that under the microscope. The trichomes mature evenly throughout the plant, top to bottom. So your get your sample anywhere that causes the least damage getting to it.
I like to harvest them when they've gone from the hazy mixed colors of clear, red, and amber. I don't wait until ALL the trichomes are amber. Harvesting herb that is a little imature give you medicine that has more of a THC buzz than waiting for the CBD's to mature which gives you more of the couch medication. It's all about what your body needs to feel it's best.
Your microscope power is probably a real good power level. I get the best view when I go all the way down to 50.
